That sounds like a lock file that didn't get cleaned up properly.
If you delete the lock file and it works again, you might want to open an
enhancment request with the Ubuntu team -- during the boot-up phase it
should be safe to delete these lock files, it could be something to do
automatically.
If you continue to have this problem and can't track down what app is
causing it, you can always add a "rm -f
/var/state/apt/lists/lock" to a startup script yourself.
